ug/ul to uM

I need help with this conversion. I have an oligo that is 0.1ug/ul (20ul total). From this I need to make a 1uM stock to use in a PCR rxn......

What is the weight of 1µmol? - This would be the amount you need in 1L. From that, you can work out how much you need in the volume you need to make up.

If this is typically the sort of work you are involved in, is there a course you can do that will help you come to grips with these dilution problems? - While we converters are always happy to help, it is always MUCH better to work them out yourself than ask unknown cyberspace people.

I'm not sure I understand what you mean by the weight of the 1uM? The oligo is 329 pmoles total. I haven't been able to find a class, but I have been looking for books that could help with this sort of thing, because I do spend quit a bit of time doing dilutions....

You need to know the molecular weight of your sample. I'm not sure what an oligo is, but if you provide that information, I can help you.

1µM means a solution that has a concentration of 1 µmol per liter.

There are 1,000,000 µmols in 1 mol.

An oligo is just a short sequence of nucleotides. The molecular weight is 6094.

In a 1M solution, you would have 6094g/L (not possible, I know)

in a 1µM solution, you would have 6094µg/L (0.006094µg/µL)

Not sure how much stock solution you want to make up, but for example, 250µL would require 0.006094 x 250 = 1.5235µg.

The volume of your original solution that contains 1.5235µg is 1.5235 ÷ 0.1 = 15.235µL

To make 250µL of 1µM solution, you use 15.235µL of solution, and 234.765µL of solvent.

If you want to use all your original solution, the total volume is
20 x 0.1 ÷ 0.006094 = 328.2µL, so add 308.2µL to your original volume.

HI THERE,

Iam a tad confused with my lab manual instructions, they are asking me to add 400 ug proteinase K to my lysed cells- Calculate what volume of proteinase K at a stock solution of 20 mg/Ml will give me 400 ug. and they have only provided 30 ul of proteinase K...so how do I calculate the volume of proteinase K to calculate.. Thanks you for your help.

400 µg is 0.4 mg
Your stock is 20 mg/mL so
0.4 mg x 1 mL/20 mg = .020 mL = 20 µL

Perhaps easier to visualize, 20 mg/mL = 20 µg/µL
You need 400 µg, dividing by 20, you need 20 µL.
